Donepezil HCl
Indications/Uses
Moderate to severe dementia of the Alzheimer's type.
Administration
May be taken with or without food: Swallow whole w/ water, do not split/crush.
Contraindications
Hypersensitivity to piperidine derivatives.
Special Precautions
Succinylcholine-type muscle relaxation exaggeration during anesth. Vagotonic effects may occur on the SA & AV nodes. Closely monitor for active or occult GI bleeding especially those at increased risk of developing ulcers, history of ulcer disease, or receiving concurrent NSAIDs. Wt loss; bladder outflow obstruction; convulsions. Patients w/ history of asthma or obstructive pulmonary disease. May impair ability to drive or operate machinery. Childn. Pregnancy & lactation.
Adverse Reactions
Drug Interactions
Metabolism is inhibited by ketoconazole & quinidine. Reduce clearance w/ known CYP2D6 inhibitor. Increased rate of elimination w/ phenytoin, carbamazepine, dexamethasone, rifampin & phenobarb. May interfere anticholinergic medications activity. Synergistic effect w/ succinylcholine, similar neuromuscular blockers or cholinergic agonists eg, bethanechol.
