Amphotericin B
Indications/Uses
Systemic fungal infections caused by Aspergillosis, Blastomycosis, Candidiasis, Coccidiodomycosis, Cryptococcosis, Histoplasmosis, Mucormycosis, Paracoccidioidomycosis & Sporotrichosis. Empirical treatment of febrile neutropenia.
Dosage/Direction for Use
Initially 0.25 mg/kg/day infusion over 2-6 hr. Patients w/ good cardiopulmonary function tolerance Initially 0.3 mg/kg/day. Patients w/ impaired cardiopulmonary function or severe reaction Initially smaller dose of 5-10 mg/day. May be increased gradually to 0.5-1 mg/kg/day. Max: 1.5 mg/kg/day. Duration: 6-12 wk or longer.
Contraindications
Hypersensitivity.
Special Precautions
Liver impairment. Nephrotoxicity. Pseudomembranous colitis, bronchiolitis.
Adverse Reactions
Headache, nausea, vomiting, diarrhea, cramping, local venous pain at inj site, hypotension, anaphylactoid, neutropenia, hypomagnesaemia, abnormal liver & renal function test.
Drug Interactions
Antineoplastic agent, corticosteroid, zidovudine, cyclosporine, digitalis glycosides, muscle relaxant, thiazides.
