Indications and Dosage
Intramuscular
Premature labour
Adult: Once premature labour is arrested through IV infusion, administer 10 mcg via IM inj 3-8 hourly. Max treatment duration: 48 hours. Dosage and treatment recommendations may vary among countries and individual products (refer to specific product guidelines).

Intravenous
Premature labour
Adult: Initially, 200-500 mcg/min via IV infusion using a syringe pump. Dose may be adjusted based on patient's response or until control is reached. Max treatment duration: 48 hours. Dosage and treatment recommendations may vary among countries and individual products (refer to specific product guidelines).

Oral
Cerebrovascular and peripheral vascular disease
Adult: 10-20 mg 3 or 4 times daily. Dosage and treatment recommendations may vary among countries and individual products (refer to specific product guidelines).
Administration
May be taken with or without food. May be taken w/ meals, milk or antacids to minimise GI discomfort.
Reconstitution
IV infusion: Dilute with an appropriate amount of infusion solution to prepare the recommended concentration. Instructions for reconstitution may vary among individual products and between countries (refer to specific product guidelines).
Contraindications
Recent arterial haemorrhage.
Special Precautions
Not intended to be given for premature labour if an infection is present. Should not be used immediately postpartum. Pregnancy and lactation.
Adverse Reactions
Significant: Maternal pulmonary oedema and fetal tachycardia (IV), severe rash.
Cardiac disorders: Tachycardia, chest pain.
Gastrointestinal disorders: Abdominal distress, nausea, vomiting.
Nervous system disorders: Dizziness.
Vascular disorders: Hypotension, flushing.

Monitoring Parameters
Monitor blood pressure, hydration (maternal), and heart rate (maternal and fetal) during the IV infusion.
Drug Interactions
Concurrent administration with other vasodilators and anti-hypertensive agents (e.g. diazoxide) may cause severe hypotension.
Action
Description:
Mechanism of Action: Isoxsuprine is a vasodilator and β-adrenergic receptor stimulator. It directly relaxes the vascular and uterine smooth muscle. It increases muscle blood flow and decreases peripheral vascular resistance. Additionally, it may also produce positive chronotropic and inotropic effects on the heart and may increase cardiac output.
Pharmacokinetics:
Absorption: Well absorbed from the gastrointestinal tract. Time to peak plasma concentration: Approx 1 hour (oral).
Distribution: Crosses the placenta.
Excretion: Mainly via urine (as conjugates). Plasma elimination half-life: Approx 1.5 hours.
