Vit D3 (cholecalciferol)
Indications/Uses
To increase the serum 25(OH)D level in patients w/ vit D deficiency (serum 25(OH)D <30 ng/mL).
Dosage/Direction for Use
Adult & childn >12 yr 1 tab once daily. Dose according to serum 25(OH)D level & not exceeding 5,000 IU daily.
Administration
Should be taken with food: Take after meals.
Contraindications
Hypersensitivity. Hypercalcemia &/or hypercalciuria. Ca nephrolithiasis, nephrocalcinosis, hypervitaminosis D.
Special Precautions
Discontinue treatment if severe allergic reaction occurs eg, swelling on the face, lips, tongue & throat, trouble swallowing, itchiness, & breathing difficulty. Patients receiving cardiac glycosides are susceptible to high Ca conc, therefore, ECG & Ca level needs to be monitored. Ca conc in plasma & urine must be monitored for patients w/ long-term treatment & patients receiving thiazide diuretics. Sarcoidosis patient needs to be monitored. Pregnancy & lactation.
Drug Interactions
Anticonvulsants eg, phenytoin, phenobarb, primidone; rifampicin; INH; glucocorticoid, Ca salts; conivaptan; bile acid sequestrant; CYP3A4 inducer; deferasirox; mineral oil; orlistat; sevelamer; tocilizumab may reduce the effects of vit D3. Cytotoxic drugs, actinomycin, imidazole disrupt vit D3 activity. Avoid concurrent use w/ multivit & mineral, calcitonin, etidronate, gallium nitrate, pamidronate, or plicamycin. Vit D3 may increase effects of Al hydroxides, cardiac glycosides, Mg salt, sucralfate, vit D analog. May decrease effects of aripiprazole, axitinib, saxagliptin.
