Ursodeoxycholic acid
Indications/Uses
Primary biliary cirrhosis. Dissolution of small to medium size radiolucent, cholesterol-rich gallstones in patients w/ a functioning gall bladder.
Dosage/Direction for Use
Adult & elderly 10-15 mg/kg/day in 2-4 divided doses.
Contraindications
Calcified & pigment gallstones, radio opaque gallstones. Non-functioning gallbladders; chronic liver & PUD. Inflammatory disease of colon & small intestine. Pregnancy.
Special Precautions
Monitor liver function parameters AST (SGOT), ALT (SGPT) & γ-GT. Reduce treatment if diarrhea occurs. Discontinue therapy in cases of persistent diarrhea. Should not be used if gallbladder cannot be visualized on x-ray images or in cases of calcified gallstones, impaired contractility of the gallbladder or frequent episodes of biliary colic. Women of childbearing potential. Should not be taken during lactation.
Adverse Reactions
Pasty stools or diarrhea. Calcification of gallstones; urticaria.
Drug Interactions
Reduced effectiveness w/ cholestyramine, charcoal & antacids. Reduced absorption w/ Al-based antacids. Counteract effectiveness by increasing cholesterol in bile w/ estrogens & clofibrate. Possible increase in ciclosporin serum conc. Decrease effectiveness of dapsone. Possible decrease on serum ciprofloxacin. Reduce peak plasma conc & AUC of Ca antagonist nitrendipine.
