Indications/Uses
Hormone receptor (HR) +ve, human epidermal growth receptor 2 (HER2) -ve advanced breast cancer in postmenopausal women not previously treated w/ endocrine therapy; HR +ve advanced breast cancer in postmenopausal women w/ disease progression following endocrine therapy; HR +ve, HER2 -ve advanced or metastatic breast cancer in combination w/ palbociclib in women w/ disease progression after endocrine therapy.
Dosage/Direction for Use
Adult female including elderly 500 mg at intervals of 1 mth, w/ additional 500-mg dose given 2 wk after initial dose, administered 1 in each buttock as 2 consecutive 5 mL slow IM inj.
Contraindications
Hypersensitivity. Severe hepatic impairment. Pregnancy & lactation.
Special Precautions
Patients w/ bleeding diatheses, thrombocytopenia or taking anticoagulant treatment. Thromboembolic events in women w/ advanced breast cancer. Dorsogluteal inj site due to proximity of underlying sciatic nerve. Potential risk of osteoporosis. May interfere w/ Ab based-estradiol assays & falsely increased levels of estradiol. Observe patients who experience asthenia when driving or operating machinery. Mild to moderate hepatic impairment. Severe renal impairment (CrCl <30 mL/min). Use effective contraception while on treatment in patients of child-bearing potential. Not recommended in childn & adolescents.
Adverse Reactions
Nausea; increased hepatic enzymes (ALT, AST, ALP); asthenia, inj site reactions. UTI; hypersensitivity reactions; anorexia; headache; VTE, hot flushes; vomiting, diarrhea; elevated bilirubin; rash; back pain.
