Pregabalin
Indications/Uses
Adult: Peripheral & central neuropathic pain. Adjunctive therapy w/ partial seizures w/ or w/o secondary generalisation. Generalised anxiety disorder (GAD).
Dosage/Direction for Use
Dose range: 150-600 mg daily given in either 2 or 3 divided doses. Neuropathic pain Initially 150 mg daily given as 2 or 3 divided doses. May be increased based on individual patient response & tolerability to 300 mg daily after an interval of 3-7 days if needed. Max: 600 mg daily after an additional 7-day interval. Epilepsy Initially 150 mg daily given as 2 or 3 divided doses. May be increased to 300 mg daily after 1 wk based on individual patient response & tolerability. GAD Initially 150 mg daily. May be increased to 300 mg daily after 1 wk based on individual patient response & tolerability. Max: 600 mg daily after an additional wk.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Diabetic patients w/ wt gain. Discontinue if angioedema eg, facial perioral upper airway swelling occur. Dizziness, somnolence, loss of consciousness, confusion, & mental impairment. Discontinue if loss of vision, visual blurring or changes of visual acuity occurs. W/drawal of concomitant antiepileptic medicinal products. Inform patients prior to starting therapy about the w/drawal symptoms. Patients w/ CHF. Treatment of central neuropathic pain due to spinal cord injury. Monitor for signs of suicidal ideation & behaviors; misuse, abuse potential or dependence. Concomitant use w/ opioid analgesics. Encephalopathy. Galactose intolerance, lapp lactase deficiency or glucose-galactose malabsorption. Renal failure. Minor or moderate influence on the ability to drive & use machines. Pregnancy & lactation. Elderly may require dose reduction due to decreased renal function. Childn <12 yr & adolescents.
Adverse Reactions
Dizziness, somnolence, headache. Nasopharyngitis; increased appetite; euphoric mood, disorientation, irritability, insomnia, decreased libido; ataxia, abnormal coordination &/ tremor, dysarthria, amnesia, memory impairment, disturbance in attention, paraesthesia, hypoaesthesia, sedation, balance disorder, lethargy; vertigo; vomiting, nausea, constipation, diarrhea, flatulence, abdominal distention, dry mouth; muscle cramp, arthralgia, back pain, pain in limb, cervical spasm; erectile dysfunction; peripheral oedema, abnormal gait, fall, feeling drunk, feeling abnormal, fatigue; increased wt.
Drug Interactions
OCs, norethisterone &/ ethinyl oestradiol. May potentiate effects of ethanol & lorazepam.
