Vinblastine sulfate
Indications/Uses
Hodgkin's disease (stage III & IV), lymphocytic lymphoma, histiocytic lymphoma; advanced stages of mycosis fungoides; advanced carcinoma of the testis; Kaposi's sarcoma & histiocytosis X. Choriocarcinoma resistant to other chemotherapeutic agents; breast carcinoma.
Dosage/Direction for Use
Adult 1st dose: 3.7 mg/m2. 2nd dose: 5.5 mg/m2, 3rd dose: 7.4 mg/m2, 4th dose: 9.25 mg/m2, 5th dose: 11.1 mg/m2. Max: 18.5 mg/m2. Childn 1st dose: 2.5 mg/m2. 2nd dose: 3.75 mg/m2, 3rd dose: 5 mg/m2, 4th dose: 6.25 mg/m2, 5th dose 7.5 mg/m2. Max: 12.5 mg/m2. Dose given once wkly.
Contraindications
Hypersensitivity. Leukopenia, bacterial infection. Pregnancy & lactation.
Special Precautions
Patients w/ hepatic or renal disorder, bone marrow depression, infections, history of muscular or neural disease, chickenpox, presence of cachexia or ulcerated areas of the skin. Perform blood tests, liver & renal function tests frequently. Manifestation of bleeding tendency & infection. Leukopenia <2,000 WBC/mm3. Do not administer intrathecally. Avoid eye contamination. Women of childbearing potential. Childn. Elderly.
Adverse Reactions
Leukopenia, thrombocytopenia, bleeding, anemia; nausea, vomiting, anorexia, dipsia, stomatitis, infection of the lip, abdominal pain, rectal bleeding, ileus, constipation, cacogeusia, indigestion; anaphylactoid syndrome (urticaria, dyspnea, angioedema); alopecia, blister, rash; paraesthesia, peripheral neuritis, loss of deep tendon reflexes, dysbasia, convulsion, arthralgia, myalgia, headache, dizziness, depression, hyposomnia, aspermia, gonad disorder; acute shortness of breath, severe bronchospasm; dysuria, flush, phlebitis, inj site pain, sphacelism, boil or wart, lymph node pain, tachycardia; cellulitis, phlebitis.
Drug Interactions
Concomitant use w/ other antineoplastic or radiotherapeutic agents. Enhanced drug action w/ erythromycin. May cause MI or cerebral infarction w/ vinca alkaloids.
