Indications/Uses
Complicated skin & skin structure (cSSS), intra-abdominal infections (cIAI) & community-acquired pneumonia caused by susceptible strains of microorganisms in patients ≥18 yr.
Dosage/Direction for Use
Adult Initially 100 mg, followed by 50 mg every 12 hr by IV infusion over approx 30-60 min every 12 hr. Duration of treatment: 5-14 days (cSSS or cIAI) or 7-14 days (community acquired pneumonia). Ped patients 12-17 yr 50 mg every 12 hr, 8-11 yr 1.2 mg/kg every 12 hr IV to max of 50 mg every 12 hr.
Contraindications
Special Precautions
Not suitable w/ hospital acquired, including ventilator-associated pneumonia & bacteremia. Anaphylactic reactions. Avoid in patients w/ known hypersensitivity to tetracyclines; patients w/ cIAI secondary to intestinal perforation. Reserve tigecycline treatment in situations when alternative treatments are not suitable. Hepatic dysfunction. Monitor LFTs. Photosensitivity, pseudotumor cerebri, anti-anabolic action, pancreatitis. Discontinue in patients suspected of developed pancreatitis & Clostridium difficile-associated diarrhea (CDAD). Permanent tooth discoloration (yellow-gray brown). Increased risk of development of drug-resistant bacteria in absence of proven or strongly suspected bacterial infection. Pregnancy (2nd & 3rd trimester) & lactation. Childn <8 yr.
Adverse Reactions
Nausea, vomiting, diarrhea. Inj site inflammation, pain, reaction, edema & phlebitis; septic shock, allergic reaction, chills; thrombophlebitis; anorexia, jaundice, abnormal stools; increased creatinine, hypocalcemia, hypoglycaemia; taste perversion; prolonged aPTT & prothrombin time (PT), eosinophilia, increased INR, thrombocytopenia; pruritus; vag moniliasis, vaginitis, leukorrhea.
Drug Interactions
Prothrombin time or other suitable anticoagulation test should be monitored if tigecycline is administered w/ warfarin. May affect pharmacokinetic properties w/ P-gp inhibitors (eg, ketoconazole or cyclosporine) or inducers (eg, rifampicin).
