Indications/Uses
Chronic HBV infection in adults w/ evidence of active liver inflammation.
Dosage/Direction for Use
Compensated liver disease Nucleoside-naïve patients 0.5 mg once daily. Patient w/ renal impairment 30 to <50 mL/min 0.25 mg once daily or 0.5 mg once every 48 hr, 10 to <30 mL/min 0.15 mg once daily or 0.5 mg once every 72 hr, <10 mL/min, on hemodialysis or continuous ambulatory peritoneal dialysis (CAPD) 0.05 mg once daily or 0.5 mg once every 5-7 days. Lamivudine-refractory patients 1 mg once daily. Patients w/ renal impairment 30 to 50 mL/min 0.5 mg once daily or 1 mg once every 48 hr, 10 to <30 mL/min 0.3 mg once daily or 1 mg once every 72 hr, <10 mL/min, or hemodialysis or CAPD 0.1 mg once daily or 1 mg once every 5-7 days. Decompensated liver disease 1 mg once daily.
Administration
Should be taken on an empty stomach: Take at least 2 hr before or after meals.
Contraindications
Special Precautions
Monitor hepatic function during therapy & closely monitor at repeated intervals for at least several mth after discontinuation. Patients w/ unrecognized or untreated HIV infection; HIV testing in all patients prior to initiation of therapy. Do not use for the treatment of HBV in HIV-infected patients who are not receiving antiretroviral therapy. Discontinue if lactic acidosis or pronounced hepatotoxicity occurs. Pregnancy & lactation.
Adverse Reactions
Headache, fatigue, dizziness, nausea, diarrhea, dyspepsia, vomiting, somnolence & insomnia. ALT elevations, hematuria, lipase elevations, glycosuria, hyperbilirubinemia, fasting hyperglycemia & creatinine increases.
Drug Interactions
Serum conc may be increased w/ drugs that reduce renal function or compete for active tubular secretion.
