Anidulafungin
Indications/Uses
Invasive candidiasis including candidemia in adults & paed patients ≥1 mth. Esophageal candidiasis in adults.
Dosage/Direction for Use
Adult Invasive candidiasis including candidemia 200 mg as single loading dose on Day 1, followed by 100 mg daily, thereafter. Continue therapy for at least 14 days after the last +ve culture. Esophageal candidiasis 100 mg as single loading dose on Day 1, followed by 50 mg daily, thereafter. Duration of treatment: Min of 14 days & for at least 7 days following resolution of symptoms. Paed patient ≥1 mth Invasive candidiasis including candidemia 3 mg/kg as loading dose (not to exceed 200 mg) on Day 1, followed by 1.5 mg/kg (not to exceed 100 mg) daily, thereafter. Continue therapy for at least 14 days after the last -ve culture.
Contraindications
Hypersensitivity to anidulafungin or other echinocandins eg, caspofungin.
Special Precautions
Discontinue use if anaphylactic reactions including shock occur. Infusion-related reactions eg, rash, urticaria, flushing, pruritus, dyspnea, bronchospasm & hypotension. Monitor for worsening hepatic function in patients who develop abnormal LFTs during therapy. Not to be given in patients w/ hereditary fructose intolerance. Hepatic dysfunction, hepatitis, or hepatic failure. Pregnancy & lactation. Not recommended in neonates <1 mth. Infants & childn <2 yr.
Adverse Reactions
Hypokalemia; diarrhoea, nausea. Clostridium colitis, candidiasis; thrombocytopenia, thrombocythaemia; hyperkalaemia, hyperglycaemia, hypomagnesaemia, hypercalcaemia, hypernatraemia; convulsion, headache; atrial fibrillation, ventricular extrasystole; HTN; bronchospasm; vomiting, constipation; cholestasis; rash, pruritus; back pain; increased ALT, AST, blood creatinine, urea, alkaline phosphatase & bilirubin, abnormal LFT, decreased blood Mg.

Increased steady-state AUC w/ cyclosporin.
