Ondansetron is a highly selective 5HT3-receptor antagonist both centrally and peripherally. Chemotherapeutic agents and radiotherapy may cause release of 5HT3 in small intestine. Then 5HT3 initiates a vomiting reflex by activating via 5HT3-receptor of vagal afferent in the area postrema and emetic center of the brain causing release of 5HT3.
