Action

Ondansetron is a highly selective 5HT₃-receptor antagonist both centrally and peripherally. Chemotherapeutic agents and radiotherapy may cause release of 5HT₃ in small intestine. Then 5HT₃ initiates a vomiting reflex by activating via 5HT₃-receptor of vagal afferent in the area postrema and emetic center of the brain causing release of 5HT₃.
