Indications/Uses
Infections due to β-lactamase-producing strains (where amoxicillin alone is not appropriate) including resp tract infections, cellulitis, animal bites & severe dental infection w/ spreading cellulitis. Short-term treatment of bacterial infections including upper resp tract (eg, ENT particularly tonsillitis, sinusitis, otitis media); lower resp tract (eg, acute exacerbations of chronic bronchitis, lobar & bronchopneumonia); GUT (eg, recurrent cystitis or complicated excluding prostatitis, septic abortion, pelvic or puerperal sepsis & intra-abdominal sepsis, pyelonephritis, female genital infections); skin & soft tissue (eg, boils, abscesses, cellulitis & wound infection); bone & joint (eg, osteomyelitis); dental (eg, dentoalveolar abscess); other infections (eg, septicemia, peritonitis).
Dosage/Direction for Use
Adult One or two 375-mg tab every 8 hr. More severe infections & infection of the resp tract One 625-mg tab every 8 hr or one 1,000-mg tab every 12 hr. Childn 25-50 mg/kg/day (equiv to amoxicillin 20-40 mg/kg & clavulanic acid 5-10 mg/kg) in divided doses every 8 hr.
Administration
May be taken with or without food: Best taken at the start of meals for better absorption & to reduce GI discomfort.
Contraindications
Hypersensitivity to β-lactam antibiotics, penicillin & cephalosporins. History of cholestatic jaundice/hepatic dysfunction. Pregnancy.
Special Precautions
Perform periodic assessment of organ system functions, including renal, hepatic & hematopoietic during prolonged therapy. Not for the treatment of sore throat or pharyngitis. Patients w/ lymphatic leukemia; severe hepatic dysfunction; moderate or severe renal impairment. Discontinue use in case of superinfection.
Adverse Reactions
Diarrhea, nausea, vomiting, indigestion, abdominal pain, skin rashes, urticaria & erythema multiforme, vaginitis, abnormal taste, headache, dizziness, tiredness & hot flushes.
Drug Interactions
Decreased renal tubular secretion of amoxicillin w/ probenecid. Prolongation of bleeding & prothrombin time w/ anticoagulants. Reduced efficacy of OCs. Potential increased incidence of skin rashes w/ allopurinol. Possible transient decreases in plasma conc of total conjugate estriol, estriolglucuronide, conjugated estrone & estradiol.
